Article Title: Tannase Production By Tannic-Acid Degrading Bacteria Isolated From Cowpea (Vigna Unguiculata)

Abstract:
This study explored the isolation and characterization of tannaseproducing bacteria from cowpea (Vigna unguiculata) seeds collected in Epe, Lagos State, Nigeria. Sixteen pure bacterial isolates, including species of Staphylococcus, Micrococcus, Pseudomonas, Enterococcus, and Bacillus, were recovered and biochemically identified. Primary screening using tannicacid agar revealed six tanninolytic isolates, with three (DVN?5, DVN?8, DVN?17A) demonstrating the largest zones of hydrolysis (up to 21?mm). Submerged fermentation over 5 days led to crude enzyme yields, with the top isolate (A1) exhibiting the highest tannase activity (32.194?U/mL). The enzyme was optimal at 60?°C, pH?9.0, and 0.8% tannic acid concentration, with magnesium (Mg²? at 1?mM) significantly enhancing activity. The study concludes that cowpea harbors diverse tanninolytic bacteria suitable for industrial tannase production under alkaline and thermophilic conditions.yme.
