| Abstract: This study investigated the ?-amylase inhibitory activities and antioxidant properties of Arachis hypogea seed protein hydrolysates. Arachis hypogea seeds were pulverized into flour, defatted using n-hexane and proteins were extracted from the resulting seed meal. The proteases pepsin and trypsin were utilized for protein hydrolysis and the hydrolysates obtained were evaluated for ?-amylase inhibitory activity and antioxidant properties (using hydrogen peroxide and ferric ions). -amylase inhibition assay showed that tryptic hydrolysates had better inhibitory activity (IC50 = 0.489±0.002mg/ml). Kinetic data indicated uncompetitive type of inhibition for both hydrolysates, with tryptic hydrolysates showing significantly higher (p<0.05) binding affinity (ki = 0.148mg/ml). Antioxidant assays indicated that hydrolyates derived from tryptic digestion showed significantly higher (p<0.05) hydrogen peroxide scavenging capacity (EC50 = 0.352±0.005mg/ml) and ferric reducing power when compared to peptic hydrolysates. These results suggest that Arachis hypogea seed protein hydrolysates appear to have multiple functionalities which could be utilized to develop newer anti-diabetic agents and food additives. |
